How do I find out which users currently have a file open?

You can right-click on the file and choose More Functions->Open By, or you can expand the server in the left window and use the Open Files option.

I have tried this with a number of files already and it doesn't seem to work. I've even had other people open the files on their machines and tested it and no user display.

Can think of any reason why this would occur?

The MIcrosoft "Open Files" function I think requires that the file be opened through a share. Hyena reports the results of this particular function (http://windowssdk.msdn.microsoft.com.../ms725129.aspx). There is also an Open Files option on the Manage Computers MMC snapin that should report the same information.
